P21

In plain English

P21 is a short peptide built from the active region of CNTF, a natural nerve growth factor, with an added chemical group that helps it reach the brain. It is studied for growing new nerve cells and protecting memory.

What it's used for

  • Memory and learning
  • Brain aging and dementia (being studied)
  • Mood

Benefits

In animal studies, P21 increased new nerve cell growth in the memory center of the brain, improved learning and memory, and reduced Alzheimer-like changes. Users report sharper memory and focus.

How it works

In simple terms: it borrows the active tip of a nerve growth factor to help the brain grow and protect neurons.

It is thought to raise BDNF and support neurogenesis in the hippocampus, while blocking the side effects of full-length CNTF. An adamantane group helps it cross into the brain.

Development history

  • 2010s: Developed by Khalid Iqbal’s group at the New York State Institute for Basic Research.

Typical protocols

Educational information only, not medical advice. The information below describes how this peptide has been used in research, on prescription labels or by users. It is not a recommendation. Talk to a qualified healthcare professional before starting, stopping or changing anything.

Doses vary; these ranges are what users commonly report.

Use Dose Frequency Length
Cognitive 200–500 mcg Daily 2–4 weeks

Cycling

Commonly run for 2–4 weeks, then a break.

Administration options

Nasal spray or subcutaneous injection.

Reconstitution and dosing calculator

Usually comes in 5 mg or 10 mg vials, mixed with 2 mL of bacteriostatic water (or saline for nasal use).

1Peptide in the vial

The total amount printed on the vial label.

2Bacteriostatic water added

More water makes each dose a bigger, easier-to-measure draw. It does not change how much peptide is in the vial.

3Dose per injection

Use the dose from your protocol or your clinician. 1 mg = 1,000 mcg.

4Your syringe

All three are U-100 insulin syringes: 100 units = 1 mL. Smaller barrels make small doses easier to read.

    Side effects and safety

    • Users report headache and mild irritability occasionally.
    • Very little safety data in people.
    • Avoid during pregnancy and breastfeeding.

    Common stacks

Semax or Selank: other nasal brain peptides.

    Storage

Powder: refrigerate or freeze. After mixing: refrigerate at 2–8 °C and use within about 3 weeks.

    Compare with

    • Cerebrolysin: a broad natural mix with human data.
    • Semax: raises BDNF too, with more human use.

    Frequently asked questions

Has P21 been tested in people?

Not in formal trials; nearly all research is in animals.
